I'm running SMF 2.0 RC3 & SimplePortal 2.3.3
Do I need to uninstall SP before upgrading SMF?

all the Mods should be uninstalled before upgrading SMF... and then reinstall all the Mods
as SP is classed as a Mod, then i would say Yes you need to uninstall it before upgrading SMF...
all of your pages and articals and such like that have been applyed through SP are in the Database so no Data should be missing once SP has been re-installed

I would just need to copy the code in some of my custom block to put them back...

I have to ask as i am having the same issue. I have installed over 60 mods on my SMF 1.1 forum but i can't uninstall all of them as some are giving errors when i uninstall them, so can i just go ahead and overwrite with the SMF upgrade package and then install all the mods, including SP, without having to uninstall them first?

you might be better of asking this question in the SMF forum...!!
if your overwriting then it would probably be better to completely delete your 1.1 version and install the full 2 version
notforgetting to take a backup of your database first.

Often, if you uninstall mods in the opposite order that you installed them, it will all go OK.
Unless, of course, you have made other changes to the SMF files in the mean time.
Another option is to use the large update package. You can use it to replace ALL of your php files, which will eliminate every change any mod has made to your files.
Your custom php blocks, which remain in the database, will be unchanged either way, and will probably work probably afterwards.
